the effects of interleukin-4 and interferon-gamma on the expression of FceR 2 on B cells



Abstract
Two types of receptors for the Fo portion of human IgE(Fc¥åR) differ in their affinity for IgE as well as in their cellular distribution and function. Type 1 Fc¥åR I) show high affinity for IgE and are detectable on mast cells and basophils.
The reaction between allergen and IgE bound to Fc¥åR I induces degranulation of basophils and mast cells, with release of chemical mediators responsible for the clinical manifestations of allergy. Type 2 Fc¥åR(Fc¥åR II) bind IgE with lower
affinity
and
are present on monocytes, macrophages, lymphocytes, platelets and eosinophils. The biding of IgE to Fc¥åR II activates a variety of effector functions in these cells.
Interleukin-4(IL-4) and interferon-¥ã(IFN-¥ã) that are secreted from the T lymphocytes influence the expression of Fc¥åR II on B cells and the production of IgE, Therefore play an important role in immunologic factors in allergy.
This study was performed to evaluate the roles of these lympokines and their interactions in the expression of Fc¥åR II and production of solubles Fc¥åR II(sFc¥åR II) on B cells.
Expression of Fc¥åR II on B cells from normal and allergic individuals were measured by double stainings with anti Leu 16-FITC and anti Lew 20-PE and analysed with FACS. Interaction between IL-4 and IFN-¥ãhydrocortisone in the expression of Fc¥åR
II
were observed.
B-lymphocytes of allergic individuals showed more increased expression of Fc¥åR II than those of % normal ones. The cultured B-lymphocytes with IL-4 showed increased Fc¥åR II expression in both groups but there was no difference between them.
IL-4
induced Fc¥åR II expression and production of sFc¥åR II were inhibited by IFN-¥ãand hydrocortisone.
Thus it may be concluded that the interaction and important role in the immunopathogenes of allergy
